RESUMO

Due to their potential release into the environment, the ecotoxicity of Ti3C2Tx (MXene) nanomaterials is a growing concern. Unfortunately, little is known about the toxic effects and mechanisms through which Ti3C2Tx induces toxicity in aquatic organisms. The aim of this study is thus to investigate the toxic effects and mechanisms of Daphnia magna upon exposure to Ti3C2Tx with different sheet sizes (100 nm [Ti3C2Tx-100] and 500 nm [Ti3C2Tx-500]) by employing conventional toxicology and metabolomics analysis. The results showed that exposure to both Ti3C2Tx-100 and Ti3C2Tx-500 at 10 µg/mL resulted in a significant accumulation of Ti3C2Tx in D. magna, but no effects on the mortality or growth of D. magna were observed. However, the metabolomics results revealed that Ti3C2Tx-100 and Ti3C2Tx-500 induced significant changes in up to 265 and 191 differential metabolites in D. magna, respectively, of which 116 metabolites were common for both. Ti3C2Tx-100-induced metabolites were mainly enriched in phospholipid, pyrimidine, tryptophan, and arginine metabolism, whereas Ti3C2Tx-500-induced metabolites were mainly enriched in the glycerol-ester, tryptophan, and glyoxylate metabolism and the pentose phosphate pathway. These results indicated that the toxicity of Ti3C2Tx to D. magna has a size-dependent effect at the metabolic level, and both sheet sizes of Ti3C2Tx can lead to metabolic disturbances in D. magna by interfering with lipid and amino acid metabolism pathways.

By separating and extracting algae from the collected water samples, an oil-producing diatom strain was obtained. Microscopic observation of the strain revealed that its morphological characteristics were highly similar to those of the genus Cyclotella. The cloning of 18S rDNA and phylogenetic analysis showed that the algae were clustered with Cyclotella menegheniana with a high support rate, indicating that the alga was C. menegheniana. The fatty acid content of the alga was determined and found to be mainly C14, C16, and C18 fatty acids, which were in accordance with the relevant standards for edible oil. In this study, different gradient levels of salinity and light were set to investigate the culture and bioactive substance production of C. menegheniana. The results showed that the best growth condition was achieved when the salinity was 15 g·L-1, and its biomass and oil content were the highest at 0.27 g·L-1 and 21%, respectively. The final biomass was the highest when the light intensity was 2000 Lux and the oil content was 18.7%. The results of the study provided a basis for the large-scale production of edible oils and biodiesel.

OBJECTIVE: To analyze the forces of rotational wall vessel (RWV) bioreactor on small tissue pieces or microcarrier particles and to determine the tracks of microcarrier particles in RWV bioreactor. METHODS: The motion of the microcarrier in the rotating wall vessel (RWV) bioreactor with both the inner and outer cylinders rotating was modeled by numerical simulation. RESULTS: The continuous trajectory of microcarrier particles, including the possible collision with the wall was obtained. An expression between the minimum rotational speed difference of the inner and outer cylinders and the microcarrier particle or aggregate radius could avoid collisions with either wall. The range of microcarrier radius or tissue size, which could be safely cultured in the RWV bioreactor, in terms of shear stress level, was determined. CONCLUSION: The model works well in describing the trajectory of a heavier microcarrier particle in rotating wall vessel.
